河南食管癌高发区AGT Exon3与食管癌高易感性关系分析

【摘要】 目的探讨AGTExon3与食管癌高易感性之间的关系。方法聚合酶链反应(PCR)法,PCR基础上的限制性片段多态(PCR-RFLP)检测法及单链构象多态性分析(PCR-SSCP)检测法检测275例食管癌患者血标本抽提DNA,以正常血标本抽提DNA315例为对照。结果AGT第三外显子(AGTExon3)突变型携带者患ESCC的危险度比野生型携带者升高4.876倍(95%CI,3.248 ̄7.319)。将AGTExon3各基因型组合考查河南安阳地区人群患ESCC的结合危险度。在ESCC组,AGTExon30R值为0.1166,性别OR值为93.558,年龄为1.0703,具有统计学显著性意义。结论AGTExon3突变型基因型显著增加河南安阳地区人群ESCC的易感性,而DNA损伤修复酶基因缺陷可能是导致食管癌变的重要途径。

【Abstract】 Objective To characterize the relation between O6-alkylguanine-DNA alky ltransferase (AGT) Exon3 and high susceptibility, to esophageal cancer at high-incidence area for esophageal cancer in Henan.Methods It was applied by Polymerase Chain Reaction (PCR) and PCR based Restriction Fragment Length Pol ymorphisms (PCR-RFLP) and single strand conformation polymorphism analysis of PCR products (PCR-SSCP).Results The sex-and age-adjusted OR for AGT Exon3 mutaion genotype indicated a 4.876- fold (95% CI, 3.248~7.319) increased risk for developing ESCC when compared with the AGT Exon3 wild type(P<0.001). Using subjects with the combination of AGT Exon3 genotypes, combined risk for developing ESCC was observed. In ESCC group, the OR values of AGT Exon3 is 0.1166. The OR values of gender and age are 93.558 and 1.0703. The difference was significantly different(P<0.001). Conclusion The mutant genotypes of AGT Exon3 polymorphisms raised risk of ESCC m the population of Anyang District in Henan, the Chinese highest incidence area for these cancers.The present results indicate that DNA repair enzymaticly genic bug is the important pass to lead to ESCC.
