三氯苯达唑对卫氏并殖吸虫同工酶、蛋白质、0核酸和糖原的影响

【摘要】 目的探讨三氯苯达唑杀灭卫氏并殖吸虫的机理。方法组织化学方法用茚三酮反应显示蛋白质;过碘酸反应显示糖原及多糖物质;Brachet法显示核酸。在用药前后虫体连续切片中的分布及含量;SDS-PAGE分析比较用药前后虫体蛋白质区带;用微量平板淀粉胶电泳法进行等位基因酶谱检测。结果药物能使虫体各种组织的蛋白质均衡而显著减少,实质组织中的糖蛋白显著减少,破坏虫体谷氨酸脱氢酶(GDH),磷酸甘露糖异构酶(PMI),乳酸脱氢酶(LDH)及黄嘌呤脱氢酶(XDH)。结论三氯苯达唑对卫氏并殖吸虫的杀灭作用主要是破坏、抑制虫体酶和蛋白质的合成和分解,而且影响虫体的核酸代谢。

【Abstract】 To explore the mechanism of triclabendazole to kill Paragonimus westermani,the levels of proteins,glycogen and nucleic acids of the flukes were determined by ninhydrin reaction,periodic acid reaction and Brachet method respectively.In addition,SDS-PAGE assay was used for protein analysis,and the micro-volume plate starch gel electrophotesis was employed for the detection of the enzyme spectrum of alleles.The experimental results showed that the protein level in various tissues of worms after treatment with triclabendazole was evenly reduced,with a remarkable reduction of glycoprotein in parenchymal tissues.The enzymes in the flukes,such as glutamic dehdrogenase(GDH),phspho-mannose isoenzyme(PMI),lactic dehydrogenase(LDH) and xanthine dehydrogenase(XDH),were all destroyed by triclabendazole.It is concluded that the killing effect of troclabendazole is mainly to destroy or inhibit the synthesis and decomposition of proteins and enzymes and to influence the metabolism of nucleic acids in the flukes.
