银杏雄株亲缘关系的AFLP分析

【摘要】 【目的】研究29个中国银杏雄株材料的亲缘关系,以期为该树种雄株的分类、保存、利用等提供理论依据。【方法】从64对EcoRⅠ/MseⅠ引物(其中MseⅠ引物为荧光标记物)中筛选出7对清晰、多态性高的引物,对来自中国的29个银杏雄株材料的亲缘关系进行了AFLP分析。【结果】7对引物共产生1173条谱带,187个特异位点和1145条多态带,多态带的比例平均为97.6%。每对引物鉴别效率达到100%。所有材料之间相似系数在0.38~0.82之间,供试材料遗传变异大。当相似系数为0.51,将全部试材可以分为2大类,山东的大部分材料聚为一类。【结论】同一产地内不同试材和不同产地的试材存在不同程度的遗传差异。供试材料的聚类与其地理位置的远近不完全相关,山东大部分银杏雄株试材亲缘关系较近,‘天目山4’、‘宁国’、‘郯城6’可能是比较原始的雄株。

【Abstract】 【Objective】In order to provide theoretical evidence for classification, preservation, and utilization of Ginkgo biloba males, genetic relationship of 29 Ginkgo biloba males in China is studied. 【Method】Seven clear and high polymorphic primer combinations were screened from 64 EcoRⅠ/MseⅠprimer combinations, and the genetic relationship of 29 Ginkgo biloba was analyzed by AFLP. 【Result】7 primer combinations generated 1 173 bands, 187 specific loci and 1 145 polymorphic bands, the percentage of polymorphic bands was 97.6%. The identification percentage per primer combination was 100%. The genetic similarity coefficient among all materials varied from 0.38 to 0.82, and their genetic variation varied largely. The 29 Ginkgo biloba males were divided into 2 types by a similar coefficient, most from Shandong clustered together. 【Conclusion】Genetic variations existed in different materials from the same resource origins as well as from different resource origins. The test materials’ cluster was not completely related to their geographical distance. Genetic relationships of most materials from Shandong were relatively close. ‘Tianmushan4’, ‘Ningguo’ and ‘Tancheng6’ might be relatively original males.
