微生物基因组DNA氯化苄提取方法优化的研究
Modified extracted method for genomic DNAs from microbe using benzyl chloride
【摘要】 以PichiapastorisGS115为例,通过正交设计对氯化苄提取基因组DNA条件进行优化,并获得最优水平组合提取条件:提取液pH9、氯化苄加量300μL、保温时间60min。以PichiapastorisGS115的基因组DNA为模板,PCR扩增泛素基因(Ubi)在琼脂糖凝胶电泳228bp处出现DNA亮带。利用该法提取其他微生物基因组DNA也能够获得很好的结果。
【Abstract】 In this paper,conditions for extracting genomic DNAs from Pichia pastoris GS115 using benzyl chloride was optimized by orthogonal test.The optimum conditions were: pH 9.0,adding 300 μl benzyl chlorides and keeping temperature for 60 min.After PCR was operated using the isolated genomic DNAs of Pichia pastoris GS115 as template and using a double of specific primers,a 228 bp segment of Ubi was obtained.
