食管癌细胞表面DR5的表达分布与细胞铺展状态的关系

【摘要】 目的:探讨食管癌鳞癌EC9706细胞系细胞表面DR5的表达分布与细胞铺展状态的关系。方法:免疫细胞化学技术分析食管癌EC9706细胞的DR5表达分布;流式细胞术检测食管癌细胞表面DR5的表达及细胞的凋亡率。结果:DR5分布在食管癌EC9706细胞的细胞质和细胞膜表面,主要在细胞质,但在细胞核没有DR5分布,而且在悬浮的食管癌细胞表面DR5表达高于铺展者。悬浮的食管癌细胞对抗DR5功能性抗体诱导的细胞凋亡敏感性高于铺展者。结论:食管癌细胞表面DR5的表达与细胞铺展状态具有密切的关系,细胞铺展状态的变化直接影响细胞表面DR5的表达分布以及对抗DR5功能性抗体诱导的细胞凋亡敏感性。该研究结果对进一步探讨DR5表达分布的机制以及TRAIL和DR5的功能性抗体的抗肿瘤临床应用有一定的指导意义。

【Abstract】 Objective:To reveal the relationship between cell spreading and DR5 distribution on the cell surfaces of human esophageal carcinoma EC9706 cell line.Methods:DR5 distribution of EC9706 cells was observed under fluorescence microscope by immunocytochemical assay. DR5 expression on EC9706 cell surfaces and EC9706 cell apoptosis were detected by flow cytometry analysis.Results:DR5 primarily was located in the cytoplasm and on the surfaces of EC9706 cells, mainly in the cytoplasm but not in the nuclei. DR5 expression on the surfaces of the floating cells was much higher than of the spreading cells, in addition, the floating cells were more sensitive to the apoptosis induced by anti-DR5 agonistic antibody mDRA-6 than the spreading cells.Conclusion:There is a close relationship between DR5 expression on the surfaces of EC9706 cells and the cell spreading, and the cell spreading maybe have an important effect on DR5 expression on the cell surfaces and on the sensitivity of the cells to the apoptosis induced by antibody mDRA-6 or TRAIL. The results maybe provide new insights into the regulation mechanisms of DR5 expression and distribution on cell surfaces, and also provide potential instruction significance for future clinical application of antibody mDRA-6 and TRAIL.
