【摘要】 目的:通过检测AIRE(AutoImmueREgulator)基因在NOD(Nonobesediabetic)鼠相关组织及1型糖尿病(Type1diabetesmellitus,T1DM)病人PBMC中的表达,从分子水平上探讨AIRE在免疫耐受中的作用。方法:运用半定量RTPCR方法检测AIRE在BALB/c鼠(n=5)及NOD鼠(n=5)相关组织及T1DM病人的外周血单个核细胞(PBMC)中的表达情况,用βactin作为内参。结果:在BALB/c及NOD鼠中,Aire主要表达在免疫系统相关组织:胸腺、脾脏及淋巴结,另外在卵巢中也有表达,并且在NOD鼠中Aire含量与BALB/c鼠相比明显减少。部分T1DM病人PBMC上AIRE表达缺失。结论:AIRE基因的表达异常与T1DM的发病具有一定的相关性。

【Abstract】 Objective:To investigate the effects of AIRE on T1DM development by detecting the expression of AIRE in NOD mice and PBMC of T1DM patients.Methods:AIRE expressions in tissues of BALB/c, NOD mice and PBMC of T1DM patients were analyzed by semi-quantitative RT-PCR, using β-actin as an internal standard.Results:In BALB/c and NOD mice, AIRE gene was predominantly expressed in immunological relevant tissues, such as thymus, spleen and lymph node. In addition, it is also expressed in ovray; But in NOD mice, AIRE gene expressions were profound reduced as compared with BALB/c mice. In some of T1DM patients, AIRE gene expressions in PBMC were defective.Conclusion:T1DM development is related with abnormal expression of AIRE gene.
