细胞免疫缺陷与认知衰老的关系研究

【摘要】 目的探讨细胞免疫缺陷与认知衰老的关系及其可能的作用机制。方法随机选取相同周龄(22~24周龄)的细胞免疫缺陷小鼠(Balb/c-nu/nu裸鼠)和正常小鼠(Balb/c)各6只,用W estern b lot法分析海马突触体素(Synaptophysin,Syn)表达水平和ELISA法检测海马促炎细胞因子IFN-γ、TNF-α含量。结果细胞免疫缺陷小鼠海马Syn表达水平显著低于正常小鼠(P<0.05),并伴有IFN-γ水平显著增高(P<0.01)。结论细胞免疫缺陷加速认知衰老过程,其机制可能与其所致的海马突触数量减少和IFN-γ增高有关。

【Abstract】 Objective To explore the relationship between deficit in cellular immune function and cognitive aging and its action mechanism.Methods The nude mice(Balb/c-nu/nu mice) and healthy mice(normal Balb/c mice) aged 22~24 weeks were selected as experimental group or control group,6 mice in each group.The expression levels of synaptophysin(Syn) in hippocampus were analyzed by Western blot and the contents of cytokines IFN-γ and TNF-α in hippocampus were measured by ELISA.Results The expression of Syn was lower significantly(P<0.05) and IFN-γ levels(P<0.01)were higher in experimental group than those in control group.Conclusions Deficit in cellular immune function may speed up the process of cognitive aging by reducing the number of synapse and increasing IFN-γ levels in hippocampus.
