脑脉通及其拆方对局灶性脑缺血大鼠自由基代谢和细胞因子的影响

【摘要】 目的从自由基代谢和细胞因子方面观察脑脉通及其拆方抗脑缺血损伤作用,探讨其抗脑缺血损伤的作用机制及其脑脉通组方配伍规律。方法采用线栓法建立大鼠局灶性脑缺血模型,检测各组大鼠脑缺血后神经症状、脑组织含水量、脑梗死面积以及脑组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量和TNF-α、IL-1β、TGF-β水平。结果与假手术组比较,模型组大鼠脑梗死面积增加,神经功能障碍加重,脑组织含水量增加,IL-1、TNF水平和MDA含量增高,SOD活性降低;与模型组比较,尼莫地平组、脑脉通及各拆方组脑梗死面积、神经功能障碍、脑组织含水量,脑组织IL-1、TNF水平和MDA含量有不同程度降低,SOD活性和TGF-β水平有不同程度的升高,其中脑脉通组效果较显著。结论脑脉通及其拆方对脑缺血损伤具有保护作用,其机制可能与其拮抗自由基损伤和调节细胞因子有关。

【Abstract】 Objective To observe the effects of Naomaitong and of its different of ingredient groups against cerebral ischemia injury on the metabolism of free radical and the level of cytokine,and to investigate the mechanism of its against cerebral ischemia injury and the rule of composition or the formula of Naomaitong.Methods The SD rats were subjected to middle cerebral artery occlusion with intraluminal filament technique,the effect of every group on nervous symptom,oedema of brain,infarct size and the activity of SOD,the content of MDA,the level of TNF-α,IL-1β,TGF-β were detected.Results Infarct size of brain were significantly greater,nervous symptom was more serious,oedema was higher,and the level of MDA,TNF-α and IL-1 was higher,the activity of SOD was lower in model group than those of sham-operated group.The nervous symptom,oedema of brain,infarct size and the level of MDA,TNF-α and IL-1 of brain in treated groups were lighter and the level of TGF-β and SOD was higher than those of the model group,especially those of Naomaitong group.Conclusions Naomaitong and its different of ingredient could protect brain damage after focal cerebral ischemia,its mechanism might be related to relieving the injury caused by free radical and regulating the disorder levels of cytokines.
