B-ALL患者胸腺输出TCR Vβ亚家族nave T细胞分析

【摘要】 目的了解B细胞-急性淋巴细胞白血病(B-ALL)患者外周血T细胞中TCR Vβ亚家族23种信号T细胞受体删除DNA环(sjTRECs)的存在情况,从而了解胸腺近期输出各TCRVβ亚家族naveT细胞的功能。方法利用半巢式PCR分别扩增10例B-ALL患者外周血单个核细胞DNA中的23种Vβ~Dβ1sjTRECs,10例正常人作为对照。结果TCRVβ亚家族sjTRECs在大部分正常人和部分B-ALL患者中均可检测到,与正常人比较,B-ALL病人的Vβ24~Dβ1sjTRECs出现频率比正常人高,且有统计学意义(80%∶30%,P=0.0285),另有18个家族的Vβ~Dβ1sjTRECs比正常人低,其中在Vβ1(30%,P=0.0285)、Vβ12(20%,P=0.0285)、Vβ13(40%,P=0.0043)、Vβ15(20%,P=0.0022)和Vβ18(20%,P=0.0089)5个亚家族sjTRECs出现频率的差异有统计学意义。结论B-ALL病人胸腺输出大多数Vβ亚家族naveT细胞功能低于正常人。

【Abstract】 Objective In order to investigate the recent thymic output function of TCR Vβ subfamily nave T cells in patients with B cell-acute lymphoblastic leukemia (B-ALL) by detection of the frequencies of 23 subfamilies of TCR Vβ sjTRECs in peripheral blood T cells.Methods 23 subfamilies of Vβ-Dβ1 sjTRECs were amplified in genomic DNA of peripheral blood mononuclear cells (PBMC) from 10 cases with B-ALL by using semi-nest PCR.10 normal individuals served as controls.Results TCR Vβ subfamilies of sjTRECs were detected in normal individuals and some cases with B-ALL. Compared with the normal control, the frequencies of Vβ24-Dβ1 sjTRECs in B-ALL patients (80%) was significant higher than in the normal individuals (30%, P= 0.0285 ), while lower frequencies were found in the other 18 Vβ subfamily sjTRECs in B-ALL. The frequencies of Vβ1-(30%, P= 0.0285 ), Vβ12-(20%, P= 0.0285 ), Vβ13-(40%, P= 0.0043 ), Vβ15-(20%, P= 0.0022 ) and Vβ18-Dβ1 sjTRECs (20%, P= 0.0089 ) subfamilies were significant lower than in the normal individuals.Conclusion The recent thymic output function of most Vβ subfamily nave T cells in patients with B-ALL was lower than in the normal individuals.
