矮化杉木蛋白质组的差异凝胶电泳分析
Proteome study of dwarf mutant in Cunninghamia lanceolata by 2D-DIGE
【摘要】 建立具有高分辨率和稳定性的矮化杉木Cunninghamia lanceolata叶片蛋白质的双向电泳图谱,研究杉木矮化突变的机理。取同一生长环境下的野生型杉木及矮化突变型杉木新梢顶端的叶片,液氮研磨成粉末后用改良TCA-丙酮沉淀法提取总蛋白,分别用CY2和CY3标记,用DIGE技术进行分析。与野生型杉木相比,在矮化突变型杉木的叶片组织中,有14个蛋白质表达水平显著增加,另外15个蛋白质表达水平显著下降。所得的29个差异蛋白质可能与杉木矮化突变的发生有关。图3表3参9
【Abstract】 To establish the maps of two-dimensional differential gel electrophoresis,the dwarfing mechanism of the mutant in Cunninghamia lanceolata was analyzed.The leaves from normal and dwarf plants were ground into powder with liquid nitrogen.The total proteins were extracted with the modified TCA-acetone method,labeled with CY2 or CY3 respectively,analyzed by 2D-DIGE.After electrophoresing,the gels were respectively imaged by Typhoon 9400 using different emission filter and spot-features were analyzed by DeCyder software in each paired normal/dwarf comparison.The results showed that 14 unique proteins the had a 2.2-fold increase and 15 proteins that had a 3.0-fold decrease were found in dwarfed plants.These 29 proteins might be associated with the dwarfing.
