大鼠心肺复苏后海马CA1区胶质纤维酸性蛋白和S100β蛋白的表达

【摘要】 目的动态观察大鼠心肺复苏后海马CAI区胶质纤维酸性蛋白(GFAP)和S100β蛋白表达的变化。方法成年雄性SD大鼠80只,随机分为对照组和复苏组,再分别按气管切开后或自主循环恢复(ROSC)后0.5、3、6、12、24 h分为5个亚组,每组8只。建立窒息型大鼠心肺复苏模型,各亚组分别于各时间点取样,以干湿比质量法测定脑组织含水量;以免疫组化方法测定海马CAI区GFAP和S100β蛋白的表达。结果与对照组比较,复苏组ROSC后各时间点海马CAI区S100β蛋白的表达随脑水含量的上升而增加(P<0.01),且两者呈正直线相关(P=0.016);复苏组ROSC后海马CAI区GFAP表达于6 h开始逐渐增加,至24 h表达量显著增加水平(P=0.003)。结论大鼠心肺复苏后海马CAI区S100β蛋白和GFAP表达均增加,但S100β蛋白增加出现早于GFAP,并且与脑水肿程度密切相关;星形胶质细胞增殖活化所致的S100β蛋白和GFAP在不同时序表达增加,可能分别与复苏后的神经元功能损伤或修复密切相关。

【Abstract】 Objective To investigate the expression of glial fibrillary acidic protein(GFAP) and S100βprotein in hippocam- pus CA1 region after cardiopulmonary resuscitation (CPR) in rats.Methods 80 Male adult SD rats were randomly divided into 2 groups:sham - operation group and CPR group.And each group was further divided into 5 subgroups respectively on the base of time (0.5,3,6,12,24 h after incision of trachea or ROSC,n = 8).Asphyxiation cardiac arrest model was used and samples was taken on each time point of subgroups.Brain water content was measured by wet -dry weight method,and the expression of GFAP and S100βprotein was measured by immunohistochemistry.Results The expression of S100βprotein in hippocampus CA1 region elevated when brain water content increased in CPR group compared with sham - operation group (P<0.01),and a positive correlation was found between them (P=0.016).The expression of GFAP in hippocampus CA1 region started to increase at 6 h after ROSC and reaced a significant difference at 24 h in CPR group compared with sham-operation group (P=0.003).Conclusion The expression of S100βprotein and GFAP increase in hippoeampus CA1 region in the early stage after CPR in rats.However the increases of S100βprotein is previous to GFAP,and which is significantly correlative with brain edema.The increase of expression of S100βprotein and GFAP at different time series resulting from astrocytes proliferate and activate may be respectively relative to disorder or restoration of neuronal function after CPR.
