光谱法研究芦氟沙星与人血清白蛋白的相互作用

【摘要】 用光谱法研究水溶液中芦氟沙星与人血清白蛋白的结合反应,分别测定了295,310 K两个不同温度下二者的结合常数KA和结合位点数n,探讨了其荧光猝灭机制;根据热力学参数确定了芦氟沙星与人血清白蛋白之间的作用力类型,以氢键或范德华力为主,并运用F ster偶极-偶极非辐射能量转移原理,测定了其相互结合时授-受体间的距离r;采用同步荧光技术考察确定芦氟沙星对人血清白蛋白构象产生一定的影响.

【Abstract】 The binding reaction between rufloxacin(RFLX) and human serum albumin(HSA) has been studied by fluorescence spectroscopy and synchronous fluorescence spectroscopy in aqueous solution(Tris-HCl buffer,pH 7.0).The results show that the fluorescence intensity of HSA at 344 nm is quenched when RFLX is added,and the quenching mechanism of fluorescence of HSA by RFLX is a static quenching procedure,and the binding constant K_a and the number of binding sites n are calculated.The interaction between RFLX and HSA is stronger and the main binding force is hydrogen bond or Van de Waals force.The binding distance r between RFLX and HSA is also obtained according to the Frster theory of non-radiation energy.The effect of RFLX on the conformation of HSA is analyzed using synchronous fluorescence spectrometry.
