弓形虫P30乳酸球菌口服免疫小鼠血清中IFN-γ、IL-2及NO含量的测定
Detection of IFN-γ,IL-2 and NO in the sera of the mice orally immunized with recombinant Lactococcus lactis expressing P30 of Toxoplasma gondii
【摘要】 目的观察弓形虫P30乳酸球菌(L.lactis/P30)口服免疫鼠血清中细胞因子IFN-γI、L-2及NO的变化情况。方法L.lactis/P30口服免疫BALB/c小鼠,以gp42乳酸球菌组和生理盐水组为对照,在第3周、第9周及第12周分别收集各实验组小鼠血清,ELISA法测定血清IFN-γI、L-2含量;酶法测定NO浓度。结果L.lactis/P30口服免疫鼠血清中IFN-γI、L-2含量均显著高于两对照组,血清中NO含量随着免疫时间延长与两对照组差异逐渐明显。结论L.lactis/P30口服免疫能有效刺激小鼠细胞因子IFN-γ和IL-2的产生及NO分子的释放。
【Abstract】 Objective To observe the changes of IFN-γ,IL-2 and NO in the sera of the mice orally immunized with recombinant Lactococcus lactis expressing P30 of Toxoplasma gondii.Methods Recombinant L.lactis/P30 was administered orally to BALB/c mice.Control groups were administered orally with gp42 L.lactis and normal saline respectively.Sera were collected from all animals at weeks 3,9 and 12 intervals.The contents of IFN-γ and IL-2 were detected by ELISA,and the contents of NO was detected by enzyme assay.Results The contents of IFN-γ and IL-2 in sera of the mice orally immunized with recombinant L.lactis/P30 were significantly higher than that of two control groups.The difference of NO in sera between L.lactis/P30 group and control groups were obviously increased with the immunized time prolonged.Conclusion Orally immunization with L.lactis carrying P30 of T.gondii can obviously stimulate the production of IFN-γ,IL-2 and NO in BALB/c mice.
