慢性日本血吸虫感染对小鼠实验性1型糖尿病的影响

【摘要】 目的探讨慢性日本血吸虫感染对小鼠实验性自身免疫性1型糖尿病发病的影响及其机制。方法19只雄性BALB/c小鼠随机分为3组:日本血吸虫感染+糖尿病造模组(A组,7只),糖尿病模型组(B组,6只)和正常对照组(C组,6只)。A组每只小鼠感染25条日本血吸虫尾蚴后9周,与B组小鼠同时腹腔注射链脲佐菌素诱导1型糖尿病,动态观察血糖变化。至12周末剖杀小鼠,留取血清,取胰腺组织切片HE染色,进行胰岛炎评分,用ELISA测定血清干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)水平。结果给药后,小鼠血糖逐渐升高,在第21d、28d两个时间点B组显著高于A组(P<0.05);A组<40%的胰岛可见淋巴细胞浸润,B组有80%的胰岛可见淋巴细胞浸润,两者比较差异有显著性(P<0.01);至感染12周末,A组小鼠血清IL-4水平显著高于B组(P<0.01);B组小鼠血清IFN-γ水平显著高于A组(P<0.05)和C组(P<0.01)。结论慢性日本血吸虫感染对小鼠实验性1型糖尿病具有拮抗作用,其机制与Th1/Th2免疫偏移有关。

【Abstract】 Objective To investigate the effect of chronic infection with Schistosoma japonicum on multiple low-dose streptozotocin (MLDSTZ) induced diabetes mellitus and its possible mechanism. Methods Nineteen male BALB/c mice were randomly divided into 3 groups: S. japonicum infection+MLDSTZ group (group A, n=7), MLDSTZ induced diabetes group (group B, n=6) and normal control group (group C, n=6). Each mouse of group A was infected with 25 cercaria, and then type 1 diabetes was induced by MLDSTZ injection in groups A and B 9 weeks post-infection. Blood glucose levels of mice from each group were observed dynamically. By 12 weeks post-infection, mice were executed and sera were collected, then their pancreases were removed to be scored on insulitis by HE staining. The IFN-γ and IL-4 levels in serum were measured by ELISA. Results Mice injected with STZ became gradually hyperglycemic. On days 21 and days 28 after first injection, the mean blood glucose levels of group A were significantly lower than that of group B (P<0.05). There were only less than 40% of islets with lymphocytic infiltration in group A, whereas 80% of islets in group B were infiltrated by massive lymphocyte (P<0.01). IL-4 levels in group A were significantly higher than that in group B (P<0.01), IFN-γ levels in group B were higher than that in group A (P<0.05) and group C (P<0.01). Conclusion Chronic infection with S. japonicum has antagonistic effects on MLDSTZ mice, which may be related to immune deviation of Th1 to Th2.
