流式细胞术在精子质量检测中的应用

【摘要】 目的研究流式细胞检测方法在精子质量评价中的应用。方法将104例男性不育患者的精子标记相应的荧光染料,用流式细胞仪检测精子的质膜完整性、染色质状态、顶体状态和线粒体膜电位等,并与精液常规检测方法进行比较和相关性分析。结果男性不育患者精子质膜完整率为(54.20±21.23)%,染色质结构分析t(37.78±22.78)%,SDt(44.24±14.58)%,COMPt(26.24±21.42)%,线粒体膜电位正常率(50.22±18.36)%,活精子的顶体率(25.27±12.08)%,相关性分析结果表明精子的线粒体膜电位与其活动率(a+b)%存在正相关性(P=0.0002)。结论流式细胞术与常规检测方法相比,不仅具有快速、精确的优点,还可以了解精子的内部结构和功能,是评价精子质量的有效方法。

【Abstract】 Objective To investigate the function of flow cytometry in sperm quality evaluation.Methods The membrane integrity,the chromatin structure,the acrosome reaction and mitochondrial membrane potential (MMP) of sperm samples from 104 infertile male patients were analyzed using fluorescence microscope and flow cytometry (FCM),and results were compared with those with the sperm routine detection methods.Results The rate of sperm membrane integrity was (54.20±21.23)%. Sperm chromatin structure assay (SCSA) showed that t:(37.78±22.78)%,SDt:(44.24±14.58)%,COMPt:(26.24±21.42) %. The rate of sperms with normal MMP was (50.22±18.36)% and the mean rate of sperms with acrosome was(25.27±12.08)%. The correlation analysis showed that MMP of sperms was positive correlated with the motility(a+b)%(P=0.0002).Conclusion The flow cytometry is more rapid and more accurate than the routine sperm detection methods in the sperm quality evaluation and is an effective method for evaluating the sperm quality,with which the structure and the function inside the sperms can also be understood.
