【摘要】 目的探讨 CD66c 及其基因 CEACAM6在成人急性白血病细胞上的表达及意义。方法体外培养 HL-60、K562、LCL721.221和 Jurkat 等急性白血病细胞,RT-PCR 检测急性白血病患者骨髓细胞和白血病细胞中 CEACAM6 mRNA 的表达,并用多参数流式细胞术检测白血病细胞及白血病患者骨髓细胞 CD66c 分子的表达。同时对199例白血病患者骨髓细胞作细胞遗传学分析,并对25例CD66c 阳性急性 B 淋巴细胞白血病（B-ALL）患者进行白血病微量残留病（MRD）分析。结果①HL-60、K562、LCL721.221和 Jurkat 细胞的细胞膜和细胞质 CD66c 均阴性。②127例急性髓系白血病（AML）患者中4例 CD66c 阳性（3.15%）,以 M₂、M₄为主;79例 ALL 中 CD66c 阳性28例（35.44%）,均为 B-ALL;CD66c 只在 common B-ALL（54例中有20例）和 pre B-ALL（11例中有8例）亚型表达,8例 PH⁺B-ALL 患者 CD66c 均阳性。③MRD 阳性与阴性组患者比较,6个月内复发率差异有统计学意义（维持治疗14周时 MRD 阳性者8例,其中6例复发;MRD 阴性者17例,其中2例复发）。④CEACAM6 mRNA 在 CD66c 阳性 B-ALL 细胞强表达,在 HL-60细胞呈弱表达,在 K562、LCL721.221和 Jurkat 细胞不表达。结论 CD66c 在 ALL 中阳性表达是白血病 MRD 检测的标志之一。CD66c 抗原表达与 CEACAM6 mRNA 的表达高度相关。更多还原

【Abstract】 Objective To explore the expression of CD66c(CEACM6)in adnlt acute leukemia and its significance.Methods Acute leukemia cell lines HL-60,K562,LCL721.221and Jurkat were cultured in vitro.RT-PCR and multi-parameter flow cytometry were applied to analysis of CD66c mRNA and protein ex- pression respectively in the cell lines and patient’s bone marrow leukemic cells.Cytogenetic analysis for 199 bone marrow samples from leukemia patients and Minimal Residual Disease(MRD)detection for 25 CD66c positive B lineage ALL were performed.Results ①CD66c expression both on cell surface and in plasma were negative in all the cell lines.②Four of 127 AML(3.15%)(mainly of M2 and M4),and 28 of 79 ALL(35.44%)(all of B linage ALL)were CD66c positive the subtypes of the ALL being common B-ALL (20/54)and pre B-ALL(8/11)including 8 Ph+B-linage ALL.③Six-month relapse rate was significantly different between the MRD positive and negative patients.④CD66c mRNA was strongly expressed in B-linage ALL.For the cell lines,only the HL60 cells weakly expressed CD66c mRNA.Conclusion CD66c expres- sion could be a useful bio-marker for the MRD analysis in ALL,and is closely associated with its transcription level.更多还原
