双酚A对人神经母细胞瘤SK-N-SH细胞增殖的影响
Effect of bisphenol A on the proliferation of SK-N-SH human neuroblastoma cells
【摘要】 目的探讨双酚A(BPA)对人神经母细胞瘤SK-N-SH细胞增殖的影响及其可能机制。方法SK-N-SH细胞经培养扩增后分为5组:组1,无干预(对照组);组2,加17β-雌二醇(E2);组3,加BPA;组4,同时加E2和ICI182,780;组5,同时加BPA和ICI182,780。分别在0h、24h、48h和72h进行光吸收度值(AV)检测,在72h进行流式细胞仪DNA增殖指数(PI)检测。结果组3的AV在24h、48h、72h均明显增加,分别是组1的1.53、1.25、1.20倍,72h时的PI是组1的1.42倍。组2的结果与组3相似。组4、5中细胞增殖受到ICI182,780的明显抑制。结论BPA能促进人神经母细胞瘤SK-N-SH细胞的体外增殖,该促增殖作用可能是通过雌激素受体依赖途径实现的。
【Abstract】 Objective To investigate the effect of bisphenol A (BPA) on the proliferation of SK-N-SH human neuroblastoma (NB) cells in vitro.Methods NB cells were cultured in estrogen-free improved minimum essential medium and divided into 5 groups based on different treatments: group 1, no treatment; group 2, treated with 17β-estradiol (E_ 2 ); group 3, treated with BPA; group 4, treated with both E_ 2 and ICI 182,780; group 5, treated with both BPA and ICI 182,780. Absorbance value (AV) was determined at 0, 24, 48 and 72 h, and DNA proliferation index (PI) was detected by flow cytometry at 72 h.Results At 24, 48 and 72 h, the AV of group 3 were 1.53 , 1.25 and 1.20 times than those of group 1, respectively. At same time, PI in group 3 was 1.42 times than that of group 1 at 72 h. The results of group 2 were similar to those in group 3. However, the growth of cells was inhibited by ICI 182,780 despite the presence of E_ 2 and BPA in group 4 or 5.Conclusions BPA can promote the proliferation of SK-N-SH cells to a level similar to that of E_ 2 , which is probably via estrogen receptor pathways.
