落新妇甙对心脏移植排斥反应中细胞因子表达的影响

【摘要】 目的探讨落新妇甙对心脏移植排斥反应中细胞因子表达的影响。方法采用小鼠颈部心脏移植模型,随机分为3组:对照组,移植组,落新妇甙组。于术后第5d取移植心脏,用逆转录聚合酶链式反应(RT-PCR)法观察IL-2、γ-IFN、IL-4、IL-10、IL-12、IL-18的表达情况。结果移植组IL-2、γ-IFN、IL-12、IL-18表达明显升高,IL-4、IL-10表达显著降低(与对照组相比,P<0.01)。落新妇甙组IL-2、γ-IFN、IL-12、IL-18表达明显降低,而IL-4、IL-10表达显著升高(与移植组相比,P<0.01)。结论落新妇甙抑制心脏移植排斥反应主要与其抑制IL-2、γ-IFN、IL-12、IL-18等Th1型细胞因子的表达,促进Th2型细胞因子IL-4、IL-10的表达,使免疫反应由Th1型向Th2型偏移有关。

【Abstract】 Objective To investigate the effects of Astilbin on the expression of IL-2, γ-IFN, IL-4, IL-10, IL-12 and IL-18 in mice heart transplantation rejection.Methods Model of mice cervical heterotopic heart transplantation was established. The mRNA of IL-2, γ-IFN, IL-4, IL-10, IL-12 and IL-18 were measured by RT-PCR at 5 days after transplantation in control, transplant and Astilbin group.Results Compared to the control group, the mRNA levels of IL-2, γ-IFN, IL-12 and IL-18 were significantly increased while those of IL-4 and IL-10 were significantly decreased in transplant group (P< 0.01 ). In Astilbin group, the mRNA levels of IL-2, γ-IFN, IL-12 and IL-18 were significantly decreased while IL-4 and IL-10 were significantly increased (P< 0.01 ) compared to the transplant group.Conclusions Astilbin can decrease the expression of IL-2, γ-IFN, IL-12 and IL-18 while increase the expression of IL-4 and IL-10 in mice heart transplantation rejection.
