肿瘤坏死因子相关凋亡诱导配体对裸鼠胃癌移植瘤的生长抑制作用与死亡相关蛋白3的关系
TRAIL inhibits gastric cancer implanted in nude mice and its relation with death associated protein 3
【摘要】 目的观察肿瘤坏死因子相关凋亡诱导配体(TRAIL)对裸鼠胃癌移植瘤的生长抑制作用,探讨死亡相关蛋白3(DAP3)与其的关系。方法建立人胃癌细胞株BGC-823裸鼠移植瘤模型;观察不同剂量的TRAIL(0、1、5、10mg/kg体重)对移植瘤的生长抑制作用;流式细胞仪测定细胞周期及凋亡情况;Western blot和逆转录-聚合酶链反应(RT-PCR)检测DAP3蛋白和DAP3 mR- NA表达情况。结果5 mg/kg体重和10 mg/kg体重两组的瘤重分别为(2.57±0.35)g和(2.49±0.26)g,均低于1 mg/kg体重组(3.23±0.32)g和对照组(3.57±0.47)g,前两组的细胞增殖指数分别为(36.69±5.16)%和(39.69±6.13)%,低于对照组(48 06±6.99)%;Western blot和RT- PCR法分别测得前两组中DAP3蛋白和mRNA的表达高于1 mg/kg体重组和对照组。结论TRAIL可有效抑制裸鼠胃癌移植瘤的生长,其作用机制可能与诱导DAP3表达有关。
【Abstract】 Objective To observe the effect of tumor necrosis factor-related apoptosis-inducing lig- and(TRAIL)on human gastric cancer BGC-823 and to evaluate its relation with death associated protein 3(DAP3).Methods Nude mice model implanted sub-cutaneously with BGC-823 was established.Dif- ferent doses of TRAIL(0,1,5 and 10 mg/kg)were administrated intraperitoneally.The rate of tumor inhibition and apoptosis of tumor were measured.The expression of DAP3 protein and mRNA was detect- ed by Western blot and RT-PCR method respectively.Results The tumor weight of the 5 mg/kg group and 10 mg/kg group was(2.57±0.35)g and(2.49±0.26)g respectively,both significantly less than that of control group.The expression of DAP3 protein and mRNA was increased in 5 mg/kg and 10 mg/ kg group.Conclusion TRAIL could inhibit human gastric cancer BGC-823 implanted in nude mice.Its mechanism might be related to enhanced expression of DAP3.
