β1整合素反义寡核苷酸对人胰腺癌BXPC-3细胞体外侵袭力的影响
Effect of integrin-β1 antisense oligodeoxynucleotides on invasive ability of pancreatic carcinoma BXPC-3 cells in vitro
【摘要】 目的观察β1整合素反义寡核苷酸(ASODN)对人胰腺癌BXPC-3细胞体外侵袭力的影响。方法采用脂质体将不同浓度β1整合素ASODN转染到人胰腺癌BXPC-3细胞中,逆转录-聚合酶链反应(RT-PCR)、Western印迹法和Transwell侵袭室方法分别检测细胞β1整合素mR- NA、蛋白表达水平及体外侵袭能力。结果与对照组相比,32~128mg/L ASODN转染后可抑制81整合素mRNA和蛋白的表达(P<0.05),64mg/L ASODN转染使BXPC-3细胞体外侵袭力明显下降(P<0.05)。结论靶向β1整合素的ASODN可有效抑制其在人胰腺癌BXPC-3细胞中的表达,并降低癌细胞体外侵袭力。
【Abstract】 Objective To investigate the effect of integrin-β1 antisense oligodeoxynucleotides(A- SODN)on invasive ability of pancreatic carcinoma cell line BXPC-3 in vitro.Methods Different concen- trations of integrin-β1 ASODN were transfected into BXPC-3 cells with liposome.The expressions of inte- grin-β1 mRNA and protein was determined by RT-PCR and Western-blot.The invasive ability was mea- sured by the number of cells to penetrate polycarbonates coated with matrigel.Results In contrast with the control group,the expressions of integrin-β1 mRNA and protein was significandy decreased after. transfected with 32-128 mg/L ASODN(P<0.05);Invasive tests in vitro also showed that the invasive ability of BXPC-3 cells was inhibited by transfection with 64 mg/L integrin-β1 ASODN.Conclusion In this cell line model,integrin-β1 ASODN could effectively inhibit the expression of integrin-β1 gene,thus inhibit the protein synthesis,and decrease the ability of invasion.
