抗人膀胱癌单克隆抗体BDI-1的扩增和鉴定
Production and characterization of antihuman bladder carcinoma monoclonal antibody BDI-1
【摘要】 目的制备纯化的抗人膀胱癌单克隆抗体BDI-1。方法给BALB/C小鼠腹腔注射BDI-1杂交瘤细胞,收集的腹水过Protein G-agaros亲和层析柱即得到人膀胱癌BDI-1单克隆抗体。采用酶联免疫吸附试验(ELISA)法、间接免疫荧光法、SDS-聚丙烯酰胺凝胶电泳法和点印迹法鉴定单克隆抗体。结果腹水可以得到1.0~1.5g/L的高效价(10~6)的纯化抗体。结论该方法可以扩增高效价的单克隆抗体。
【Abstract】 Objective To prepare purified antihuman bladder carcinoma monoclonal antibody BDI-1.Methods BALB/C mice were subjected to the intraperitoneal injection of hybridoma BDI-1.By using protein G-agaros affinity chromatography,antihuman bladder carcinoma monoclonal antibody BDI-1 was obtained from the collected ascites.Monoclonal antibody was characterized by ELISA,indirect im- munofluoresence,SDS-PAGE electrophoresis and dot blot.Results 1-1.5 mg high titer(10~6)and puri- fied antibody was obtained from ascites.Conclusion This method can produce high titer monoclonal an- tibody.
