全反式维甲酸对脑胶质瘤细胞Cyclin E、Caspase-3表达的影响
Effects of all-trans retinoic acid on expression of Cyclin E,caspase-3 in glioma cell
【摘要】 目的探讨全反式维甲酸对大鼠C6脑胶质瘤细胞增殖抑制作用及细胞周期素Cy- clin E和凋亡相关基因Caspase-3 mRNA表达的影响。方法噻唑蓝(MTT)比色法检测全反式维甲酸(ATRA)在不同浓度(1、2、4、8、16μmol/L)、不同作用时间(24、48、72 h)处理大鼠胶质瘤C6细胞系后,对细胞增殖抑制率的变化;逆转录-聚合酶链反应(RT-PCR)法检测细胞周期素Cyclin E和Caspase-3 mRNA在24、48、72 h不同时间点表达变化。结果ATRA对C6细胞有增殖抑制作用,具有时间和浓度依赖性;药物作用组与无药物作用组之间及药物作用各组之间的OD值和抑制率比较差异有统计学意义(P<0.01)。随ATRA作用时间的延长,细胞周期素Cyclin E mRNA表达逐渐下降,与细胞凋亡相关基因Caspase-3 mRNA表达逐渐增高(P<0.01)。结论ATRA抑制C6脑胶质瘤细胞发生增殖,其机制可能与细胞周期素Cyclin E表达下调,凋亡相关基因Caspase-3上调有关。
【Abstract】 Objective To study the effects of all-trans retinoic acid on cell proliferation and ex- pression of the cyclin E,Caspase-3 in rat C6 glioma ceils.Methods C6 glioma cells were cultured in vitro with 1,2,4,8 and 16μmol/L ATRA for 24,48,72 h respectively.The rate of inhibition was examined by MTT.The expression of cyclin E and Caspase-3 was detected by RT-PCR.Results MTT showed that ATRA could suppress the proliferation of C6 glioma cells in a dose-and time-dependent manner. There was significant difference in A values and the inhibition rate between ATRA experimental groups and control group(P<0.01).The expression of cyclin E mRNA was down-regulated and that of Cas- pase-3 mRNA up-regulated after ATRA treatment(P<0.01).Conclusion ATRA can inhibit the pro- liferation of rat C6 cells probably by down-regulating cyclin E and up-regulating Caspase-3 gene.
