杂色曲霉素对人食管鳞状上皮细胞人类白细胞抗原Ⅰ分子表达的影响

【摘要】 目的探讨杂色曲霉素(ST)对体外培养的人原代食管鳞状上皮细胞人类白细胞抗原I(HLA-I)蛋白表达的影响。方法从手术切除的正常食管组织分离食管黏膜鳞状上皮,立即进行人食管鳞状上皮细胞的原代培养。以不同浓度ST(2、1、0.5、0.25、0.125 mg/L)处理24 h。采用免疫细胞化学(IHC)和流式细胞分析(FCM)方法分析ST对体外培养的人食管上皮细胞HLA-I 表达的影响。结果 IHC和FCM结果均表明,经不同浓度ST处理24 h后,较高浓度ST(0.5、1、2 mg/L)组食管鳞状上皮细胞HLA-Ⅰ分子表达明显下降(P<0.05)。在0.125 mg/L到2 mg/L浓度范围内,随ST处理浓度的升高,HLA-Ⅰ表达逐渐降低,两者呈明显的负相关。结论在0.125 mg/ L到2 mg/L浓度范围内,ST可抑制人食管上皮细胞HLA-Ⅰ分子的表达,在一定程度上影响食管鳞状上皮的抗原递呈功能。

【Abstract】 Objective To study the effects of of sterigmatocystin (ST) on the expression of HLA-I in the in vitro cultured human primary esophageal epithelial cells. Methods Normal human esophageal mucosa was obtained from fresh surgically resected specimens. Immediately after removal, the esophageal squamous epithelial cells were isolated and cultured in vitro. The cultured esophageal squamous epithelial cells were treated with different concentrations (2.0,1.0, 0.5, 0.25 and 0.125 mg/L) of ST for 24 h. HLA- I expression was detected by immunocytochemistry and flow cytometry. Results Im-munocvtochemsitry and flow cytometry revealed that the expression of HLA-I in esophageal squamous epithelial cells in vitro after ST treatment for 24 h at higher concentration groups (0.5,1. 0, 2.0 mg/L) was significantly lower than in control group ( P < 0.05 ). A dose-effect relationship between ST concentration and the inhibitory effect on the HLA- I expression was found at the concentration range from 0.125 mg/L to 2.0 mg/L. Conclusion Within the dosage range from 0.125 mg/L to 2.0 mg/L, ST could inhibit the expression of HLA- I in esophageal squamous cells in vitro dose-dependently, and thus affect the antigen presenting function of the treated cells.
