自发性癫癎大鼠脑内电压门控钠通道亚型mRNA的研究
The study of voltage-gated sodium channel subtype mRNA in spontaneously epileptic rat brain
【摘要】 目的对照研究自发性癫癎大鼠(SER)和正常Wistar大鼠(WTC)脑内Ⅰ、Ⅱ、Ⅲ型钠通道α亚基mRNA表达情况。方法提取枕叶皮质、齿状回及海马CA1和CA3区组织总RNA,应用逆转录-聚合酶链反应(RT-PCR)及建立限制性内切酶图谱检测Ⅰ、ⅡA、ⅡN、ⅢA和ⅢN型钠通道的表达。结果SER枕叶皮质、齿状回及海马CA1和CA3区总钠通道(3.179±1.967、3.009±2.364、3.071±2.036、1.440±0.761)的表达略高于WTC(2.112±1.454、1.474±1.257、2.198±0.978、1.399±0.853),但差异均无统计学意义(P>0.05),SERⅢ型钠通道在海马的表达(1.066±0.276)高于WTC(0.419±0.098),且P<0.01,酶切图谱显示ⅢN型钠通道表达水平有明显上调。结论SER脑内海马Ⅲ型电压门控钠通道N亚型表达上调。
【Abstract】 Objective To compare the expression of coding voltage dependentⅠ,ⅡandⅢsodium channelα-isoforms in spontaneously epileptic rats(SER) and control rats. Methods The entire RNA was extracted from occipital cortex,dentate gyrus, CA1 and CA3 hippocampus,all types of sodium channels were observed using PCR A protocol after reverse transcription,and the expression of typeⅠ,ⅡA, IⅡN,ⅢandⅢA,ⅢN sodium channels was respectively determined after restricted digestion of PCR B,C and D products. Results In SER,all types of sodium channel expressed in a higher level (3.179±1.967, 3.009±2.364,3.071±2.036 and 1.440±0.761,respectively) than those of control groups in occipital cortex (2.112±1.454),dentate gyrus (1.474±1.257),CA1(2.198±0.978) and CA3 hippocampus (1.399±0.853) without significant differences (P>0.05). Relative proportion of sodium channelⅠ,ⅡA,ⅡN andⅢmRNAs in four adult brain areas was not significant different between control rats and SER (P>0.05). But typeⅢsodium channel expressed higher than that in control groups in hippocampus, restriction mapping analysis showed thatⅢN increased significantly (P<0.01). Conclusion The expression of typeⅢsodium channelⅢN is up regulated in hippocampus of SER brain.
