司他夫定的合成
Synthesis of Stavudine
【摘要】 β-胸苷和对甲苯磺酰氯在吡啶中反应制得的3~+,5~■-二(对甲苯磺酰基)胸苷,在LiOH作用下醚化后,在KOH-t-BuOH体系中进行β-消除反应制得司他夫定,总收率69%。
【Abstract】 Stavudine was synthesized fromβ-thymidine by reaction with p-tosyl chloride in pyridine with DMAP as catalyst to give 3’,5’-di-p-tosyl thymidine,which was subjected to etherification in the presence of LiOH followed byβ- elimination in KOH-t-BuOH system with an overall yield of 69%.
