β-环糊精包合技术在微生物转化生产氢化可的松中的应用
Application of β-Cyclodextrin Inclusion Technique in the Biotransformation of Hydrocortisone
【摘要】 为提高甾体生物转化的底物投料浓度,采用超声法制备底物RS-β-环糊精包合物。以新月弯孢霉为实验菌种,培养后加入溶有包合物的磷酸盐缓冲液制备氢化可的松。结果表明,采用β-环糊精包合后甾体底物浓度可由2g/L提高至3g/L,2L发酵罐中氢化可的松收率为57.8%。
【Abstract】 Inclusion compound RS-β-cyclodextrin (RS-β-CD) was prepared by ultrasonication and used to increase the concentration of steroid substrate in the biotransformation of hydrocortisone by Curvularia lunata in phosphate buffer. The result showed that the concentration of steroid substrate could be increased from 2g/L to 3g/L, and the yield of hydrocortisone was 57.8% in 2L fermentor.
