麻黄素对体外培养的人鼻腔纤毛运动的影响

【摘要】 目的观察不同浓度麻黄素对体外培养的人鼻腔纤毛运动的影响。方法采用组织块培养法,观察在加有羟乙基哌嗪乙磺酸[4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid,HEPES]的Hank平衡盐溶液(Hank balanced salt solution,HBSS)、2.5 g/L、5g/L、10g/L和20g/L的麻黄素作用下体外培养人鼻腔纤毛摆动频率(ciliary beat frequency CBF),并进行配对分析。结果①CBF在HBSS中10 min内无显著性变化;②在加入2.5 g/L、5 g/L、10 g/L和20 g/L的麻黄素后,1~2 min内CBF明显升高并达高峰,与加药前相比差异有统计学意义(P值均<0.05),后渐呈下降趋势,在10 min时2.5g/L和5g/L的麻黄素中CBF与加药前相比虽略低,但差异无统计学意义(P值均>0.05),而10 g/L和20 g//L麻黄素中CBF与加药前相比明显降低,且差异有统计学意义(P值均<0.05);③2.5 g/L~20 g//L的麻黄素作用1 min后CBF的增加量差异无统计学意义(F=0.697, P>0.05),10 min后随着浓度的增加而下降,10 g/L~20 g/L麻黄素CBF的下降值较2.5 g/L~5g/L的下降值差异有统计学意义(P<0.05)。结论麻黄素作为肾上腺素受体激动药物,2.5 g/L~20 g/L的麻黄素对体外培养的人鼻腔纤毛运动早期有一定的刺激作用,后期则表现为抑制作用,且随着药物浓度的增加,刺激作用逐渐缩短,抑制作用逐渐延长。5g/L麻黄素作用于鼻腔纤毛细胞刺激作用最大,而抑制作用较弱。

【Abstract】 Objective To investigate the effects of ephedrine on human nasal cilia movement. Methods Ciliary beat frequency ( CBF) of cultured human nasal epithelial cells was measured by highspeed digital microscopy in HBSS and ephedrine solution of different concentrations in 10 minutes. Results CBF of cultured nasal epithelial cells exposed to HBSS showed no significant changes in 10 minutes. However, in 2. 5 g/L , 5 g/L, 10 g/L and 20 g/L ephedrine solution, CBF increased significantly in 1-2 minutes and reached the apex, then it decreased gradually, at the 10th minute. CBF of the samples exposed to 2. 5 g/L and 5 g/L ephedrine solution were slower than those in HBSS, but no significant changes were found. However, in 10 g/L and 20 g/L ephedrine solution, CBF decreased significantly when compared with samples in sHBSS. With the concentrations from 2. 5 g/L to 20 g/L ephedrine, the increment was independent on the concentration, the inhibitory effect was dependent on the concentration. Conclusions In initial time, 2. 5 g/L-20 g/L ephedrine stimulated CBF, then 10 g/L-20 g/L ephedrine inhibited CBF. The stimulation of 2. 5 g/L and 5 g/L ephedrine on CBF was longer than that of 10 g/L and 20 g/L ephedrine. 5 g/L ephedrine had maximum stimulatory effect without obvious inhibitory effect on cultured human nasal CBF.
