局部注射BMP-2对老年大鼠股骨上端骨密度和骨强度变化的影响

【摘要】 目的观察局部注射骨形态发生蛋白(BMP2)后老年大鼠股骨上端骨密度及骨强度的变化。方法22月龄大鼠10只,同一个体双侧对照,股骨上端以Pluronicf127为载体局部注射BMP2,对照侧仅注入Pluronicf127,术后4周、8周处死取材,应用CT扫描、双能X线骨密度测定及生物力学测试来观察局部皮质厚度、骨密度及骨强度的变化。结果4周时,试验侧与对照侧局部皮质厚度、骨密度及骨强度无明显差异;8周时,试验侧骨密度及骨强度较对侧明显增高(P<0.05和P<0.01)。结论局部注射BMP2可显著提高老年大鼠股骨上端局部皮质厚度、骨密度及骨力学强度,其可能是预防老年性骨质疏松骨折的一种有效方法。

【Abstract】 Objective To study the effect of BMP-2 injection intramedullarily on bone density and strength of the proximal femur in aging rats. Method Ten rats,22 months old,served as experimental model with BMP-2 and pluronic f-127 injected intramedullarily in the right upper femur.The left femur of the same rat was taken as control with pluronic f-127 injection only.Specimens were obtained at the 4 th week and 8 th week after the injection when rats was sacrificed.Thickness of the cortex,bone density and strength were tested by CT,Dual-energy X-ray absorptiometry(DEXA) and biomechanical test. Result At the 4 th week,no difference was found in the BMD,bonestrength and the thickness of cortex between the two sides,while an increase was shown in the right femur at the 8 th week. Conclusions BMP-2 injection intramedullarily can improve the cortex thickness,bone density and strength of the proximal femur in aging rats and may be effective in preventing senile osteoporosis fractures.
