红曲对体外培养成骨细胞增殖、分化及矿化功能的影响
Effects of Hongqu onthe Proliferation,Cell Differentiation and Mineralization of the Osteoblast Cultured in Vitro
【摘要】 目的:研究中药红曲对体外培养成骨细胞增殖、分化及矿化功能的影响。方法:取SD大鼠48只,随机将大鼠分为A组(空白组)、B组(倍美力组)、C组(普拉固组)、D、E、F组(红曲高、中、低剂量组)六组,按10 ml/kg分别予以生理盐水、倍美力水溶液(6.25μg/ml)、普拉固水溶液(0.2 mg/ml)及红曲水提液(1.25g/ml、0.625 g/ml、0.125 g/ml)灌胃。灌胃10天后,制备含药血清;将从乳鼠颅骨取材的成骨细胞培养于含药血清培养液中,然后应用MTT法、对硝基苯磷酸盐法及茜素红染色方法观察体外培养成骨细胞、碱性磷酸酶及矿化结节的变化。结果:经过10天红曲水提液灌胃后采血制备的含药血清明显增加了成骨细胞数量(P<0.01或P<0.05)、碱性磷酸酶的表达水平及矿化结节的形成,作用随红曲剂量的增加而增加。结论:成功地运用血清药理学的方法,模拟了体内的药理环境;证实了红曲含药血清可以呈剂量依赖性地促进成骨细胞增殖。
【Abstract】 Objective :To study the effects of Hongqu on the proliferation,cell differentiation and mineralization of theosteoblast cultured in vitro.Methods :Take 48 SD rats were divided randomly into 6 groups : A group(blank group),Bgroup(Premarin see conjugated estrogen group),C group(Provastain group),D,E,F group(each are high,mediumandlow dose group).Each group was given normal sodium,Premarin solution(6.25 μg/ ml).Provastain solution(0.2 mg/ml),and Hongqu extraction(1.25g/ ml,0.625g/ ml,0.125g/ ml)through intragastric administration.10 days later,killedand took the rats’blood to prepare medication-containing serum.Meanwhile,the skull osteoblast of the rats were cul-tured in serumculture-medium,and then used the method of MTT,para-nitro-pheneye phosphate and the tinctorialmethod of alizarin Bordeaux to observe the effect of Hongqu on such aspects as the proliferation,ALP expression,andmineralization tuberculation of the Osteoblast.Results :Hongqu medicine serumcould obviouslyincreace the quantity of os-teoblast(P<0.01 or P<0.05),the level of the ALP expression and mineralization tuberculation whichis obviouslyi m-proved by the close of Hongqu.Conclusions :Making use of the method of the serumpharmacology successfully,si mulatingthe pharmaco-peristasis in the body,we find out that Hongqu can present the ground of dose dependent to promote theosteoblast’s proliferation.
