高效液相色谱法测定大山楂丸和山楂鲜果中齐墩果酸及熊果酸的含量

【摘要】 目的:建立测定大山楂丸及山楂鲜果中齐墩果酸和熊果酸含量的高效液相色谱法。方法:色谱柱:Hypersil BDS C18(4.6mm×200mm,5μm);柱温为25℃;检测波长为210nm。流动相为甲醇-磷酸水溶液(pH3.0,90∶10),流速为0.5mL·min-1。结果:齐墩果酸的线形范围12.5~220μg·L-1,回归方程为A=16.86C+43.96,r=0.9993,回收率为99.98%,RSD为2.69%(n=6);熊果酸的线形范围36.0~600μg·L-1,回归方程为,A=15.78C+67.40,r=0.9994,回收率为101.1%,RSD为2.78%(n=6)。结论:该方法简便、快捷、准确,重复性好,适合于大山楂丸及山楂鲜果中齐墩果酸和熊果酸含量测定。

【Abstract】 OBJECTIVE To develop an HPLC method for the quantitative analysis of oleanolic acid and ursolic acid in Dashanzhawan pills and fresh haw.METHODS Oleanolic acid and ursolic acid were analyzed on a Hypersil BDS C_ 18 conlum and detected at 210 nm. The conlum temperature was 25 ℃ .The mobile phase consisted of methanol-water(adjusted to pH = 3.0 with H_ 3 PO_ 4 ) (90∶10) at a flow rate of 0. 5 mL·min~ -1 . RESULTS Calibration graphs were constructed in the range of 12.5 ng/μ1~220 ng/μL for oleanolic acid and 36.0 ng/μ1~600 ng/μl for ursolic acid. The linear correlation equations were A= 16.86 C+ 43.96 (r= 0.9993 ) for oleanolic acid and A= 15.78 C+ 67.40 (r= 0.9994 ) for ursolic acid, respectively. The average recoveries were 99.98% (n=6) for oleanolic acid and 101.11% for ursolic acid. The RSD of the method was below 3.0% (n=6).CONCLUSION This methods is fast, reliable, accurate and suitable for the determination of oleanolic acid and ursolic acid in Dashanzhawan pills and fresh haw.
