半边旗二萜类化合物5F体外抗支气管哮喘小鼠气道纤维化的作用

【摘要】 目的:观察半边旗二萜类化合物5F对体外培养的支气管哮喘小鼠气道成纤维细胞增殖和胶原合成及结缔组织生长因子(CTGF)表达的影响。方法:将不同浓度(8,32,128mg.L-1)的5F作用于体外培养的支气管哮喘小鼠气道成纤维细胞,通过噻唑蓝比色法(MTT)检测5F对细胞增殖的作用;逆转录聚合酶链反应(RT-PCR)检测其对细胞中CTGF mRNA表达水平的影响;化学比色法检测其对细胞胶原蛋白含量的影响。结果:半边旗5F对支气管哮喘小鼠气道成纤维细胞的增殖具有显著的抑制作用,还能显著降低胶原蛋白的含量(P<0.01);细胞中的CTGF mRNA的表达水平显著下调,并且均呈现量效的依赖关系。结论:半边旗5F具有体外抗支气管哮喘小鼠气道纤维化的作用,这为寻找有效治疗哮喘的药物提供了新的思路。

【Abstract】 OBJECTIVE To observe the effect of 5F on the collagen synthesis and the expression of CTGF in asthmatic mice of airway fibroblast cultured in vitro.METHODS Different concentrations of 5F(8,32,128 mg·L-1)were used to treat the cells.Thiazole bule colorimerg was used to detect the effect of 5F on cell proliferation;RT-PCR to detect the expression of CTGF mRNA;Chemical colorirnetry to detect the content of collagen protein of cells.RESULTS There were significant depress function of 5F on cell proliferation and the content of collagen protein was depressed significantly too(P<(0.01)).The expression of CTGF mRNA depressed strikingly and both showed the dependent relation with dose.CONCLUSION It shows that 5F has the function of antifibrosis in vitro,which provides a new way of finding effective drug for asthma.
