【摘要】 目的:建立测定胆汁中环丙沙星浓度的高效液相色谱分析方法,研究环丙沙星在胆管残石患者体内的胆汁药动学。方法:胆汁样品经液-液提取后,酸化,取水层进样。以乙腈-枸橼酸缓冲液(18∶82,v/v)为流动相,经色谱分离测定。结果:测定胆汁样品中环丙沙星的线性范围为0.05~10.0mg.L-1,定量下限为0.05mg.L-1。3例胆管残石患者静脉滴注环丙沙星(400mg)后,12h平均胆汁药物排泄量为0.057%。结论:本方法操作简便,灵敏度高,适用于临床药动学研究。更多还原

【Abstract】 OBJECTIVE A rapid and sensitive HPLC method for the analysis of ciprofloxacin in human bile was developed and the pharmacokinetics of ciprofloxacin in intra and extra hepatic bile duct stone patients was studied.METHODS Biliary sample containing ciprofloxacin and I.S.with liquid-liquid extraction,acidation and aqua layer to inject.Acetonitrile-citric acid buffer（18∶82,v/v）（adjusted to pH 3.0 with triethylamine）was served as mobile phase,followed by liquid chromatographic separation.RESULTS The calibration curve for ciprofloxacin in human bile was linear in the range of 0.05 to 10.0 mg·L^-1.The method was applied to the measurement of ciprofloxacin concentration in human bile.Following intravenous administration 400 mg dose ciprofloxacin to intra and extra hepatic bile duct stone patients.The excretion of ciprofloxacin in human bile was（0.088%） in 12 h.CONCLUSION The method is showed to be accurate,robust and convenient,and suitable for clinical pharmacokinetics of ciprofloxacin.更多还原
