丝裂霉素磁性前体药物纳米粒的制备

【摘要】 目的:筛选磁性葡聚糖-丝裂霉素前体药物的制备工艺,并初步考察其性质。方法:利用正交试验设计考察了制备所需的pH值、药物与载体葡聚糖的重量比、药物与催化剂的重量比以及反应浓度等对接枝率、载药量以及粒径的影响。结果:在pH5.0~5.5时,药物与葡聚糖重量比为1.5∶10,1-乙基-3-(3-二甲基氨丙基)-碳二亚胺(EDC)与葡聚糖重量比为2∶1的条件下,所得到的接枝率可达到91.7%,载药量达到8.57%,粒径平均为135nm,跨距小于0.30。结论:通过正交设计得到了高接枝率和载药量的磁性前体纳米药物。

【Abstract】 OBJECTIVE To optimize the preparation technology of the prodrug-magnetic dextran-mitomycin nanoparticles.METHODS The effects of the factors were reviewed by the orthogonal-test method including the reaction pH,the scale of the reactant and the concentration of the solution.RESULTS The highest grafting coefficient and loading capacity were occurred at the pH(5.0)-(5.5),the ratios of the mitomycin and catalyzer to the dextran were(1.5)∶10 and 2∶1 respectively by weight.CONCLUSION The diameter of the nanoparticles of the result ant product was about 49 nm,and a high grafting coefficient reached to (91.7%) and loading capacity reached to(8.57%) respectively.
