HPLC法测定布洛芬与盐酸伪麻黄碱血药浓度

【摘要】 目的建立布洛芬和盐酸伪麻黄碱血药浓度HPLC测定方法。方法采用蛋白沉淀法处理血样,以萘普生为内标,测定布洛芬血药浓度。采用液-液萃取法,以硝基苯胺为内标,测定盐酸伪麻黄碱血药浓度。测定了5名志愿受试者服药后布洛芬和盐酸伪麻黄碱血药浓度。结果布洛芬在0.5~120 mg·L-1内线性良好,回归方程为:Y=7.770 6ρ+1.2224,r=0.9988,日内日间RSD为2.57%,6.02%;盐酸伪麻黄碱在20~4000μg·L-1内线性良好,回归方程为:Y=1180.163 0ρ+5.5970,r= 0.9984,日内、日间RSD为5.33%,6.62%。结论本方法灵敏、结果准确,可供临床血药浓度检测和药动学研究使用。

【Abstract】 OBJECTIVE To establish a HPLC methods for the determination of ibuprofen and psendoephedrine hydrochloride in human plasma.METHODS The precipitation of protein was used to treat the ibuprofen sample with the internal standard of naproxen.The liquid- liquid extration was also used to treat the pseudoephedrine hydrochloride sample with internal standard para-nitroaniline.The drug concentra- tions in plasma were determined after ibuprofen and pseudoephedrine dispersible tablets were orally administered to 5 healthy volun- teers.RESULTS The calibration curve of ibuprofen was linear within the rang of 0.5~120 mg·L-1,and the regression equation was Y= 7.770 6ρ+1.222 4(r=0.998 8),the limit of quantitation(LOQ)was 0.5 mg·L-1,the average RSD of intra-day and inter-day validation were 2.57%,6.02%.The calibration curve of pseudoephedrine hydroehloride was linear within the rang of 20~4000μg·L-1,and regression equation was Y=1 180.163 0ρ+5.597 0(r=0.998 4),the LOQ was 20μg·L-1,the average RSD of intra-day and inter-day variations were 5.33%,6.62%.The blood concentrations of ibuprofen and pseudoephedrine hydrochloride were determined.CONCLUSION The sim- ple and sensitive HPLC methods are developed for the determination of ibuprofen and pseudoephedfine hydrochloride in human.
