rsTRAIL诱导人肺癌细胞系A549的细胞凋亡表达谱变化研究
Study on Apoptosis Gene Array in Lung Cancer Cell Line A549 Treated by rsTRAIL
【摘要】 目的研究一定量的重组可溶性TRAIL作用于人肺癌系A549细胞培养物后,细胞凋亡基因表达谱变化。方法用50μg·L-1的rsTRAIL处理人肺癌系A549细胞培养物,以未经药物处理的A549细胞培养物为对照,应用基因表达谱芯片技术进行细胞凋亡相关基因mRNA表达差异分析。结果经TRAIL处理后,所检测的96个细胞凋亡相关基因中,21个基因表达上调,17个基因表达水平下调。结论经TRAIL处理后,通过细胞凋亡相关的功能基因组群基因表达的正负调控,对人肺癌系A549细胞凋亡信号传导途径进行调控。药物处理不影响p53基因的表达变化。
【Abstract】 OBJECTIVE To determine the expression changes of related apoptosis genes of the lung cancer cell line A549 treated by re- combinant rsTRAIL.METHODS 50μg·L-1 rsTRAIL was added in the cultured cells,untreated culture cells were used as the control.Cells were harvested for mRNA extraction,then the genes array was analyzed.RESULTS In 96 analyzed apoptosis genes,the expression of 21 genes was up-regulated.At the same time,the expression of the other 17 genes was down-regulated.After being treated by TRAIL.CONCLUSION TRAIL regulates the apoptosis related functional genes for the regulation of the apoptosis signal transduction of the lung cancer cells.The p53 gene is not changed in the treatment.
