藻酸双酯钠对大鼠局灶性脑缺血再灌注后脑环境的影响及对鸡胚背根神经节的促生长作用
Effect of Polysaccharide Sulfate on Brain Environment after Focal Cerebral Ischemia Reperfusion in Rat and Its Promotion for Growth of the Chick Embryonic Dorsal Root Ganglion
【摘要】 目的观察藻酸双酯钠(PSS)对大鼠局灶性脑缺血再灌注后脑环境的影响及对培养的鸡胚背根神经节的促生长作用。方法采用大鼠大脑中动脉结扎模型研究PSS对局灶性脑缺血再灌注后脑环境的影响;采用鸡胚背根神经节体外培养模型研究PSS对培养的鸡胚背根神经节的促生长作用。结果在大鼠局灶性脑缺血再灌注损伤模型中,PSS可呈剂量依赖性阻止大脑皮质Ca2+释放,降低脑组织Na+,NO,丙二醛(MDA)浓度,抑制一氧化氮合酶(NOS),增强超氧化物歧化酶(SOD),Na+-K+- ATPase,Ca2+-ATPase活性,提高K+浓度;在鸡胚背根神经节体外培养模型中,PSS可呈剂量依赖性促进鸡胚背根神经节生长。结论 PSS能改善大鼠局灶性脑缺血再灌注后的损伤脑环境,促进鸡胚背根神经节生长,对局灶性脑缺血有保护作用。
【Abstract】 OBJECTIVE To investigate the effect of polysacchariole sulfate( PSS) on brain environment in rats after focal cerebral ischemia reperfusion and its promotion for the growth of the chick embryonic dorsal root ganglion(DRG) .METHODS The effect of PSS on focal cerebral ischemia was examined in rats model with middle cerebral artery occlusion (MCAO) induced by reversibly inserting a nylon thread method. The promoting growth of the DRG was determined by a culture model in vitro. RESULTS In the rats model with focal cerebral ischemia, PSS prevented Ca2+ releasing in the brain cortex, reduced the contents of Na+ , NO, MDA, inhibited the activity of NOS, enhanced the activity of SOD,Na+-K+-ATPase,Ca2+-ATPase and increased the content of K+ in the cerebrum dose-dependently. In the culture model in vitro ,PSS promoted the growth of DRG dose-dependently.CONCLUSION PSS could improve brain environment of ischemia reperfusion and promote the growth of the DRG. It implys that PSS is able to protect cerebrum against focal ischemia.
