补肾调经方对雄激素致无排卵大鼠卵巢转化生长因子β1及其受体表达的影响

【摘要】 目的:观察补肾调经方对雄激素致无排卵大鼠卵巢转化生长因子β1/转化生长因子βⅠ型受体(TGF-β1/TGF-βRⅠ)表达的影响,探讨其治疗无排卵性疾病的作用机制。方法:建立雄激素致无排卵大鼠模型,设乌鸡白凤口服液为阳性对照药,分为正常组、模型组、阳性药对照组和中药治疗组,采用HE染色观察卵巢组织形态学变化,采用免疫组化法检测卵巢颗粒细胞、间质细胞TGF-β1/TGF-βRⅠ的表达强度。结果:补肾调经方可改善病理状态下模型大鼠卵巢的形态结构,使模型大鼠卵巢颗粒细胞高表达的TGF-β1、间质细胞高表达的TGF-β1/TGF-βRⅠ恢复接近正常。结论:补肾中药可通过对卵巢TGF-β1及其受体TGF-βRⅠ表达的影响,促进卵泡发育。

【Abstract】 Objective: To observe the effect of Bushen Tiaojing recipe on the expressions of transforming growth factor-β1(TGF-β1)and TGF-β receptor type Ⅰ(TGF-βRⅠ) in the ovary of androgen-induced sterile rats(ASR) and explore the mechanism of Bushen Tiaojing recipe in treating anovulatory disease.Methods: ASR model was established by injecting androgen.Forty rats were randomly divided into 4 groups: normal control group,ASR model group,Wuji Baifeng oral liquid group(positive control group),and Bushen Tiaojing recipe group.The morphology of the ovary was observed after HE staining.The expressions of TGF-β1/TGF-βRⅠ in the ovary were detected by immuno-histochemical method.Results: Bushen Tiaojing recipe improved the ovarian morphology in pathological condition and returned the high TGF-β1 expression in ovarian granular cells and the high TGF-β1/TGF-βR1 expressions in ovarian interstitial cells to nearly normal levels in ASR.Conclusion: Bushen Tiaojing recipe,a kidney nourishing Chinese herb,may facilitate the follicular development by regulating the ex-pressions of TGF-β1/TGF-βR1 in the ovary.
