【摘要】 目的分析口服Ag85ADNA疫苗对小鼠肠上皮内淋巴细胞(iIEL)FasLmRNA表达的影响。方法减毒伤寒杆菌运载的Ag85ADNA疫苗经口免疫小鼠,检测iIEL和脾淋巴细胞FasLmRNA的表达。结果末次免疫后8d,Ag85ADNA质粒组小鼠iIELFasLmRNA平均表达水平高于正常对照组和空质粒对照组;空质粒对照组和正常对照组的iIELFasLmRNA平均表达水平以及3组脾淋巴细胞FasLmRNA的表达水平均未见显著性差异。结论口服Ag85ADNA疫苗可以上调小鼠iIELFasLmRNA的表达,而对脾淋巴细胞未见有意义的影响。

【Abstract】 Objective: To analyze the effect of oral Ag85A DNA vaccine on FasL mRNA expression in the intestinal intraepithelial lymphocytes (iIEL) of mice.Methods: The expressions of FasL mRNA in both iIEL and spleen lymphocytes were determined after oral immunization to mice using Ag85A DNA vaccine carried by attenuated Salmonella typhimurium.Results: On day 8 after last immunization, the expression of FasL mRNA in iIEL in Ag85A plasmid group was higher than that in both normal control group and empty plasmid control group. There was no significant difference in FasL mRNA expression in iIEL between normal control group and empty plasmid control group, and there was no significant difference in Fasl mRNA expression in spleen lymphocytes among these 3 groups. Conclusion: Oral Ag85A DNA vaccine could upregulate the expression of FasL mRNA in iIEL.
