细胞因子诱导的杀伤细胞对Raji细胞的杀伤效应及其机制的实验研究

【摘要】 目的探讨细胞因子诱导的杀伤(CIK)细胞对Raji细胞的杀伤效应及其作用机制。方法采用MTT法测定CIK细胞对Raji细胞的杀伤效应,用免疫组化测定bcl2蛋白的表达。结果随着CIK细胞和Raji细胞效靶比例增加及时间的延长,CIK细胞明显增强其对Raji细胞的杀伤活性,与对照组相比差异有显著性(P<0.01),bcl2蛋白的表达随时间的延长而下降,与对照组相比有显著性差异(P<0.01)。结论CIK细胞对Raji细胞有较强的杀伤活性,杀伤机制可能与下调bcl2蛋白的表达有关。

【Abstract】 Objective To explore the cytotoxicity activity and mechanism of cytokine-induced killer (CIK) cells for Raji cell lines.Methods Cytotoxicity activity of CIK cells was determined with MTT and the expression of bcl-2 protein was evaluated by immunohistochemistry.Results The cytotoxicity of CIK cells to Raji cell lines enhanced with addition of Effect/Target rate and extension of time(P<0.01),the expression of bcl-2 protein in CIK experiment group inclined when time was continued (P<0.01).Conclusion CIK cells augment killer activity to Raji cells,and the mechanism might relate to low expression of bcl-2 protein.
