【摘要】 目的:探讨川陈皮素(5,6,7,8,3′4-′hexamethoxyflavone)促非小细胞肺癌A549细胞凋亡的作用。方法:以不同浓度的川陈皮素作用于A549细胞,分别用细胞生长曲线、集落形成抑制实验研究川陈皮素对A549的增殖抑制作用,Hoechst 33258荧光染色观察细胞核形态变化,DNA琼脂糖凝胶电泳检测细胞凋亡,流式细胞术观察细胞周期改变。结果:生长曲线提示川陈皮素对A549细胞的抑制作用呈明显的时效和量效关系;集落形成抑制实验表明:药物浓度10μg/mL~40μg/mL对A549集落形成抑制率为10%~50%。Hoechst 33258染色后细胞、细胞核出现明显的凋亡形态学变化;DNA琼脂糖凝胶电泳出现典型DNA梯状条带。流式细胞仪检测到细胞周期阻滞于G2/M期,G0/G1期细胞明显减少;随着剂量的增加凋亡率明显增高。结论:川陈皮素可以诱导非小细胞肺癌A549细胞凋亡。更多还原

【Abstract】 Aim:To investigate the inhibitory effect of nobiletin（5,6,7,8,3′4′-hexamethoxyflavone） on human non-small cell lung cancer cell line A549 and its mechanism of action.Methods: The inhibitory effect of nobiletin on the proliferation of A549 cells was evaluated by the growth curve and the clone-forming assay following application of nobiletin to the cell line.The possible apoptosis-inducing effect was assessed by the morphological observation under microscope（Hoechst 33258 fluorescent staining）,flow cytometric analysis and agarose gel electrophoresis.Results: It was found that the growth curve of the cell line indicated the dose-effect and time-effect relationships.Estimation of the inhibiting rate of nobiletin to A549 in clone-forming assay ranged from 10% to 50% when nobiletin solution of ranging from 10 μg/mL to 40 μg/mL was applied.The characteristic morphology typical for apoptosis was indicated under the microscopic observation.DNA zone indicative of the apoptosis was shown on agarose gel electrophoresis.The cell cycle was arrested in G₂/M phase meanwhile it was found that cells in G₀/G₁ phase decreased and the percentage of apoptosis increased as dose size increased.Conclusion: Nobiletin could inhibit the growth of A549 cellsin vitro,and its mechanism might probably be associated with the apoptosis.更多还原
