原花青素对佐剂性关节炎大鼠腹腔巨噬细胞iNOS表达的影响
Effects of proanthocyanidins on the expression of induced nitric oxide synthase(iNOS) gene in peritoneal macrophages of rats with adjuvant arthritis
【摘要】 目的:研究原花青素对佐剂性关节炎大鼠腹腔巨噬细胞诱导型一氧化氮合成酶(iNOS)基因表达的影响。方法:建立大鼠佐剂性关节炎(AA)模型。硝酸还原酶法测大鼠关节液中NO的含量;RT-PCR和Western blot分别检测大鼠腹腔巨噬细胞内iNOS mRNA和蛋白的表达;同时ELISA检测大鼠血清中细胞因子IL-1β、TNF-α的含量。结果:原花青素显著减低AA大鼠足跖炎症组织中NO的含量以及血清中IL-1β、TNF-α的水平,同时下调AA大鼠腹腔巨噬细胞iNOS mRNA和蛋白的表达。结论:原花青素对大鼠佐剂性关节炎的抗炎作用可能与其降低TNF-α和IL-1β的水平,抑制iNOS的mRNA和蛋白表达从而减少NO的释放有关。
【Abstract】 Aim:To study the effects of proanthocyanidins on the expression of iNOS gene in peritoneal macrophages of rats with adjuvant arthritis(AA).Methods:The adjuvant arthritis model of rats was induced by the application of complete Freunds adjuvant.Contents of NO in paws of rats were measured by enzyme reduction of nitrate.The expression of iNOS mRNA and protein in rat′s peritoneal macrophages was analyzed by RT-PCR and Western blot.Contents of TNF-α and IL-1β in the rat blood serum were measured by ELISA.Results:Proanthocyanidins significantly suppressed the contents of NO in edema paws as well as serum levels of TNF-α and IL-1β of rats suffered from adjuvant arthritis.Proanthocyanidins also inhibited the expression of iNOS mRNA and protein in peritoneal macrophages of AA rats.Conclusion:The mechanisms of proanthocyanidins against adjuvant arthritis in rats is likely related to inhibition of the release of NO TNF-α and IL-1β as well as the expression of iNOS gene.
