氮酮对双氯芬酸钠透皮吸收影响的研究
Effect of Azone on Transdermal Absorption of Diclofenac Sodium
【摘要】 目的研究氮酮对双氯芬酸钠透皮吸收的促进作用。方法将BALB/cA裸小鼠和KM小鼠离体皮肤进行预处理,均分为空白组和氮酮组,分别固定于双室透皮扩散装置上进行体外透皮吸收实验。于不同时间取样,测定接收液中双氯芬酸钠的浓度,计算累积透过量、透皮速率、滞后时间。结果加用促渗剂氮酮后,双氯芬酸钠的透皮速率极显著增加(P<0.01),BALB/cA裸小鼠和KM小鼠的氮酮组分别比空白组增加2.51、2.37倍,而氮酮组透皮吸收的滞后时间均比空白组明显缩短(P<0.05)。结论氮酮对双氯芬酸钠透皮吸收有一定的促进作用。
【Abstract】 OBJECTIVE:To study the effect of azone on the transdermal adsorption of diclofenac sodium.METHODS:The isolated skin of both BALB/cA nude mice and KM mice were divided into blank group and azone group,respectively and were fixed on double-room transdermal dispersoi for the in vitro transdermal absorption tests after pretreated with azone.Samples were taken at different times for the determination of diclofenac sodium in the receiving liquid and calculations of cumulative transit dose,trandermal rate and lag time.RESULTS:The transdermal rate of diclofenac sodium increased significantly after pretreatment with azone,a kind of penetrating agent(P<0.01),increased by2.51-fold in BALB/cA nude mice(azone group)and2.37-fold in KM mice(azone group)as compared with their blank groups.The transdermal absorption lag time were shortened significantly in azone groups as compared with their corresponding blank groups(P<0.05).CONCLUSION:Azone could promote the transdermal absorption of diclofenac sodium to a certain degree.
