干扰素-γ对妊娠早期大鼠下丘脑中胰岛素样生长因子-Ⅰ表达及妊娠结局的影响

【摘要】 将妊娠9 d的大鼠随机分为对照组、试验1组、试验2组,采用生殖道肌肉注射的方法分别注射生理盐水、2.5×104U干扰素-γ(IFN-γ)、7.5×104U IFN-γ,采用免疫组织化学SP法研究IFN-γ对妊娠早期大鼠下丘脑中胰岛素样生长因子-Ⅰ的表达和妊娠结局的影响。结果显示,IGF-Ⅰ在下丘脑视前室周核、视上核、视前大细胞核、视前内侧核、视前外侧核均具有较多数量的阳性神经元表达;阳性细胞呈圆形、卵圆形、三角形,阳性物质大多位于细胞质和细胞核;生殖道肌肉注射IFN-γ后能下调IGF-Ⅰ在下丘脑的表达。同时,观察到试验2组妊娠大鼠子宫黏膜表面有大量紫褐色吸收斑。提示IFN-γ对妊娠早期大鼠IGF-Ⅰ在下丘脑的表达具有调节作用,且外源注射7.5×104U的IFN-γ后,会造成妊娠早期大鼠流产。

【Abstract】 In order to investigate effects of IFN-γ on the expression of IGF-Ⅰ gene in hypothalamus of pregnant rats and on their pregnant results,earlier pregnant rats were divided into the control group,experimental group 1 and experimental group 2.The control group was given with physiological brine and the two experimental groups were injected with 2.5×104 U of IFNγ and 7.5×104 U of IFN-γ by vaginal muscular,respectively.In results,IGF-Ⅰ were found in nucleus pre-opticus medianus,nucleus supra-(opticus,) nucleus pre-opticus mango cellularis,nucleus pre-opticus medialis and nucleus pre-opticus lateralis of hypothalamus.Positive neurons were round,oval round and triangle,which were distributed mainly in plasma and nucleus after injection.The expression of IGF-Ⅰ gene in the hypothalamus was down-regulated,and a lot of purplish red dots were observed in the uterus membrane of the pregnant rats in the experimental group 2.The results suggested that the IFN-γ could regulate the expression of the IGF-Ⅰ gene in the hypothalamus,and 7.5×104 U of IFN-γ might cause abortion in earlier periods.
