葡萄酒中11种酚酸的反相高效液相色谱测定方法研究
Studies on the Determination of 11 Phenolic Acids in Wines by Reverse Phase High Performance Liquid Chromatography
【摘要】 采用LiChrospher100RP-18e色谱柱(250mm×4.0mmID,5μm),甲醇-乙酸-水溶液为流动相,梯度洗脱,流速为1.0mL/min,SPD-6AV紫外检测器,30℃柱温,检测波长280nm。建立了一种新的、同时测定葡萄酒中11种酚酸含量的高效液相色谱法,并测定了5种不同国产葡萄酒中11种酚酸的含量。测定结果表明,葡萄酒中的酚酸主要以没食子酸、龙胆酸、咖啡酸、p-香豆酸、丁香酸等5种酚酸的含量较高,且干红葡萄酒中6种对羟基苯甲酸类酚酸总含量、5种对羟基肉桂酸类酚酸总含量和11种酚酸总含量远高于其在干白葡萄酒中的含量;不同葡萄酒中11种酚酸含量亦不相同。
【Abstract】 Five different wines made in China were used as materials,a new method for simultaneously determination of 11 phenolic acids in wines by reverse phase high performance liquid chromatography(RP-HPLC)was studied.Chromatographic separation was performed by LiChrospher 100RP-18e(250 mm×4.0 mm ID,5 μm)using the mobile phase gradient elution of methanol-acetic acid mixture at 30 ℃,flow rate at 1.0 mL/min and detection wavelength at 280 nm.The contents of 11 phenolic acids in wines of different manufactures were also determined.The results showed that the main phenolic acids in wines were gallic acid,gentisic acid,caffeic acid,p-coumaric acid and syringic acid,compared with dry white wines,the contents of total hydroxybenzoic acids,total hydroxycinnamic acids and total phenolic acids were obviously higher in dry red wines;the contents of 11 phenolic acids in different wines were different.
