鲨鱼鱼皮、鱼骨胶原蛋白的纯化及其特性的初步研究

【摘要】 胶原蛋白有许多优良性质且用途广泛。水产动物下脚料如皮、骨、鳍等含丰富的胶原蛋白。本文研究了灰星鲨鱼皮、骨胶原蛋白的提取及纯化方法。SDS-PAGE电泳分析结果显示,所提取的胶原蛋白纯度均较高。通过酶解性质的研究发现:灰星鲨鱼皮、骨胶原蛋白均可被胰蛋白酶和蛋白酶K水解,水解产物相对分子质量大小各异,成分复杂。酶解蛋白图谱结果提示:鲨鱼皮、骨胶原蛋白一级结构相近。氨基酸组成分析表明,灰星鲨鱼皮及骨胶原蛋白中,甘氨酸含量最高,其余依次是丙氨酸、谷氨酸和精氨酸。

【Abstract】 Collagens are attracting attentions recently because of its unique characteristics. A large amount of collagen exists in the wastes of aquatic animals such as in fish skin, bone and fin. Therefore, studies on collagens of fish skin and bone will offer essential values for collagen utilization. In the present study, the extraction and purification of collagens from skin and bone of shark (Mustelus griseus)were studied. SDS- PAGE revealed that the extracted collagenswere of high purity. Both collagens from skin and bone contained α1, α2 and βsubunits and they were similar to each other and they could be degraded by trypsin and proteinase K, and the hydrolyzed products were of different molecular weights. Peptide mapping suggested that the primary structures of both skin and bone collagens from the same species were of high similarity. Amino acid compositions analysis revealed that glycine was the most abundant amino acid residue in both skin and bone collagens, with alanine the second, glutamic acid the third and arginine the fourth.
