射频能量处理时间对软骨细胞及基质影响的实验研究
Experimental study on effects of radiofrequency energy treatment′s time on matrix of fibrillated articular cartilage
【摘要】 目的观察射频消融能量不同处理时间与关节软骨损伤效应之间的关系。方法建立膝关节软骨退变模型,模拟膝关节镜环境,射频消融汽化仪在同一能量设置条件下处理牛膝关节退变软骨,按不同的处理时间分为对照组、10s组、30s组、60s组4组,每组6份标本,共24份标本。随后,每份标本一分为三,分别行软骨组织块培养,HE染色,FDA/PI荧光染色,测出GAG释出率。结果随着射频消融处理时间的增加,软骨细胞空泡率明显增加,GAG释放率明显减少,软骨细胞死亡率明显增加。结论射频消融处理时间与关节软骨细胞及基质的损伤效应之间呈正相关关系。
【Abstract】 [Objective] To study on the effects of radiofrequency energy time of treatment on chondrocytes and matrix of fibrillated articular cartilage. [Methods] Setting up pattern of the bovin knee′s mechanically traumatized cartilage. In a setting designed to mimic an arthroscopic procedure. The 10 secs, 30 secs and 60 secs were treated respectively by 40W radiofrequency energy. The untreated explants served as control which contains 4 groups with 6 samples in each group and 24 samples in all, each explant was divided into 3 parts and undergone HE stain, flurescein diacetate and propidium iodide stain, the release of glycosaminoglycans. [Results] From HE stain: with the increase of radiofrequency energy treatment′s time, the death of Chondrocytes increases significantly, results from flurescein diacetate and propidium iodide stain: with the increase of radiofrequency energy treatment′s time, the death of Chondrocytes increases significantly, with the increase of radiofrequency energy treatment′s time, the release of glycosaminoglycans decreases significantly. [Conclusion] With the increase of radiofrequency energy treatment′s time, damages of the matrix and Chondrocytes of fibrillated articular cartilage was significantly servere. The damages of the matrix and Chondrocytes and the time of the radiofrequency energy treatment was positive correlation.
