ω-3多不饱和脂肪酸对人胃癌BGC823细胞周期及凋亡的影响
Effect of Omega-3 Polyunsaturated Fatty Acids on Cell Cycle and Apoptosis of Human Gastric Cancer Cell Line BGC-823
【摘要】 目的利用人胃癌BGC823细胞体外培养实验,观察不同浓度ω3多不饱和脂肪酸(EPA、DHA)对肿瘤细胞周期及凋亡的影响。方法采用MTT法、凋亡形态学检查和流式细胞检测肿瘤细胞周期及凋亡情况。结果30和45μg/ml的EPA或DHA可显著降低肿瘤细胞存活率(P<0.001);Hoechst33258染色和透射电镜可见细胞凋亡现象;流式细胞检测bcl2基因蛋白表达明显下降(P<0.05),G0/G1细胞明显增加,G2/M和S期细胞明显减少(P<0.05)。结论ω3多不饱和脂肪酸可诱导人胃癌BGC823细胞凋亡和阻滞细胞在G0/G1期,细胞增殖受到抑制。
【Abstract】 Objective To assess the effects of the ω-3 polyunsaturated fatty acids(different concentration of EPA and DHA)on cell cycle and apoptosis of human gastric cancer cell line BGC-823 in vitro. Methods Cell cycle and apoptosis of the cells were determined by MTT assay,morphology,and flow cytometry. Results After treatment with 30 and 45μg/ml of EPA or DHA,the cell viability significantly decreased(P<0.001). Apoptosis on the treated cells was observed with Hoechst33258 staining and transmission electron microscope. Bcl-2 protein expression significantly decreased and cells at G_ 2/M phase and S phase were significantly decreased (P<0.05),whereas G_ 0/G_ 1 phase was increased (P<0.05). Conclusion ω-3 polyunsaturated fatty acids can induce apoptosis of human gastric cancer cell line BGC-823 cells and inhibit the growth of the cultured cells with them accumulating in G_ 0/G_ 1 phase.
