【摘要】 目的观察补阳还五汤buyang huanwu(补阳还五)decoction,BYHWD对大鼠缺血再灌注视网膜是否具有保护作用。方法用升高眼内压的方法制作大鼠实验性视网膜缺血再灌注损伤模型。将30只大鼠随机分成对照组、缺血再灌注组和BYHWD治疗组。再灌注3天后,用体视学方法测量视网膜形态学改变,用末端脱氧核苷酸缺口标记和免疫组化方法观察视网膜细胞凋亡及bcl-2表达,比较观察BY-HWD对大鼠缺血再灌注视网膜的影响。结果BYHWD组视网膜损伤明显轻于缺血再灌注组,视网膜厚度改变不明显,内核层细胞稠密,视网膜节细胞排列较规整。BYHWD组大鼠视网膜节细胞层未见明显的凋亡细胞,bcl-2表达明显增强,阳性反应集中在内界膜处和视网膜神经节细胞周围。结论补阳还五汤复方可通过调节凋亡基因bcl-2的表达,阻断损伤后视网膜节细胞的凋亡;补阳还五汤对大鼠缺血再灌注视网膜损伤具有保护作用。更多还原

【Abstract】 OBJECTIVE To observe the effect of buyang huanwu(补阳还五)decoction(BYHWD)on the retinal ischemia-reperfusion(RIR)in rats.METHODS Retinal ischemia was induced in SD rats by increasing the intraocular pressure to 110mmHg for 60 minutes via cannulation to the anterior chamber.30 rats were divided into control group,ischemia-reperfusion group and treated group with BYHWD,randomly.Three days after reperfusion,the morphometry of retina was measured by stereologic methods;Apoptotic cells of retina were detected by TUNEL;Immunohistochemistry staining were used for the localization of bcl-2 in retina.RESULTS Of the BYHWD groups,the thickness of retina wasn′t obviosly changed.The apoptosis was less when compared with the ischemic-reperfusion groups,the apoptotic cells show only in INL and the apoptosis of retinal ganglion cells was not appeared.The bcl-2 immunoreactivity was stronger than that of the ischemic-reperfusion groups too.CONCLUSIONS Retinal apoptosis can be inhibited,and bcl-2 immunoreactivity of the retinal ischemia-reperfusion can be promoted by BYHWD.BYHWD could reduce the retinal ischemia-reperfusion injury,showing a protective effect.更多还原
