运动训练对大鼠脑缺血再灌注后功能恢复及VEGF表达的影响

【摘要】 目的:研究运动训练能否促进大鼠脑缺血再灌注后的功能恢复并探讨其机制。方法:30只SD大鼠随机分为对照组(n=10)、假手术组(n=10)和运动组(n=10),制作大脑中动脉阻断(MCAO)再灌注模型,运动组再灌注24h起每天滚笼运动训练30min,分别在再灌注12h和21d测试行为功能,免疫组化法观察VEGF蛋白的表达。结果:再灌注12h,运动组和对照组的神经功能评分、前肢放置实验和后肢放置实验评分进行比较差异无显著性意义(P>0.05),再灌注21d,运动组行为功能评分优于对照组,比较差异有显著性意义(P<0.05);运动组再灌注12hVEGF蛋白的表达水平与对照组比较差异无显著性意义(P>0.05),再灌注21dVEGF蛋白表达水平运动组明显高于对照组(P<0.05)。结论:运动训练促进大鼠脑缺血再灌注后的肢体功能恢复,其机制可能与VEGF表达上调有关。

【Abstract】 Objective:To study the effect of exercise training on the functional recovery in rats with cerebral ischemia reperfusion, and explore its mechanism.Method:30 male SD rats were randomly divided into a control group(n=10),a sham-operation group(n=10) and an exercise group(n=10). Rats were subjected to middle cerebral artery occlusion(MCAO). After 24h of reperfusion, the exercise group was trained by running wheel for 30minutes daily for 21d,other groups were without any treatment. The animals were given behavioral test and immunohistochemistry to assess functional outcome and the expression of VEGF after 12h and 21d of reperfusion respectively.Result:After 21d there was improvement in the score of behavioral test in the exercise group compared with the control(P<0.05), and the immunostraining showed that expression of VEGF was higher in the exercise group than that in the control(P<0.05).However, there was no difference between the exercise group and the control group in the score of behavioral test or in VEGF immunostraining after 12h of reperfusion.Conclusion:The exercise training can improve functional recovery in rats with cerebral ischemia reperfusion, the function enhancement may be partially contributed to the up regulation of VEGF.
