妥泰对围产期缺氧缺血性脑损伤Wistar大鼠海马少突胶质细胞的影响
The effect of Topamax on hippocampal oligodendrocyte of perinatal hypoxic-ischemic brain damage in wistar rat
【摘要】 目的观察妥泰对围产期缺氧缺血性脑损伤Wistar大鼠海马少突胶质细胞的影响。方法夹闭妊娠Wistar大鼠子宫血管,制成围产期急性缺氧缺血性脑损伤模型,治疗组于生后30min内通过胃管给予妥泰,分为1次给药组及5次给药组,观察脑缺血后再灌注7d、14d、21d、28d MBP标记的少突胶质细胞的变化。结果HI对照组新生鼠出生后21d、28d MBP阳性细胞数显著低于正常对照组(P<0.05);妥泰1次治疗组21d、28d MBP阳性细胞数显著高于HI对照组(P<0.05);妥泰5次组14d、21d、28d MBP阳性细胞数显著高于HI对照组,7d、14d、21d显著高于妥泰1次组(P<0.05)。结论妥泰可以明显减少围产期缺氧缺血性脑损伤大鼠成熟少突胶质细胞的死亡及脱髓鞘反应,且多次给药组作用强于单次给药组。
【Abstract】 Objective To investigate the effect of topamax on hippocampal oligodendrocyte of perinatal wistar rat suffered with cerebral anoxia and cerebral ischemia.Methods The perinatal cerebral anoxia and cerebral ischemia model was established by the ligation of both uterine arteries of full-term pregnant in wistar rats.Topamax was infused to stomach of rats within 30 minutes after birth.Treatment groups included administering drug once and drug five time group.Topamax effects were observed by the change of hippocampal oligodendrocyte labeled with MBP at different time spots (postnatal 7d,14d,21d,28d).Results The oligodendrocyte labeled with MBP in HI group were remarkably less than control on 21d,28d after birth(P<0.05). The oligodendrocytes labeled with MBP in administering drug once group remarkably exceeded than that in HI group on 21d,28d after birth(P< 0.05 ).The oligodendrocyte labeled with MBP in administering drug five time group remarkably exceeded than those in HI group on 14d,21d and 28d after birth,and remarkably exceeded than those in administering drug once group on 7d,14d,21d after birth(P<0.05).Conclusion Topamax can remarkably lessen the death of matured oligodendrocyte and demyelination of new-born Wistar rat suffered with cerebral anoxia and cerebral ischemia in perinatal.And it turns to be better with the increase of administration frequency.
